Yes, you can. Diploma which you have done for 3 years makes you equivalent to a 12th pass out, in fact, now if you go for B.Tech, you'll have to do it only for 3 years and not for 4 years.

A Diploma recognised by AICTE or a state board of technical education of at least 3-year duration [only for admission to IITs], you should have that then you can appear for JEE Mains. I think that class 12th is mandatory for applying to Mains, as details are required to be filled in the form. So, you cannot.
